Co-IP and immunoblotting were performed on nuclear extracts of sub-confluent cultures of the STAV-AB cells, stably transfected with syndecan-1/EGFP construct or EGFP vector, as described in “Materials and Methods”. Nuclear extract (100 ug) was incubated with 3 ug of syndecan-1 antibody, or with no antibody (negative control). The nuclear precipitates from equal amounts of control and sample proteins were slot-blotted onto a nitrocellulose membrane and then probed with antibody to FGF-2. A crude nuclear extract from syndecan-1/EGFP transfected cells was used as positive control. Co-IP using a specific antibody to syndecan-1 also pulled down FGF-2, as compared to the negative control.
