Figure 1. Western blot analysis of Talin 1 using anti-Talin 1 antibody (A02859-1). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: rat lung tissue lysates, . Lane 2: mouse lung tissue lysates, . Lane 3: human U-87MG whole cell lysates,. Lane 4: human MDA-MB-231 whole cell lysates,. Lane 5: human HepG2 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Talin 1 antigen affinity purified polyclonal antibody (Catalog # A02859-1) at 0.5 ?g/mL overnight at 4?C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Talin 1 at approximately 270KD. The expected band size for Talin 1 is at 270KD.

The tissue section was then incubated with 2?g/ml rabbit anti-Talin 1 Antibody (A02859-1) overnight at 4?C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37?C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

背景 :
Talin-1 is a protein that in humans is encoded by the TLN1 gene. This gene encodes a cytoskeletal protein that is concentrated in areas of cell-substratum and cell-cell contacts. The encoded protein plays a significant role in the assembly of actin filaments and in spreading and migration of various cell types, including fibroblasts and osteoclasts. It codistributes with integrins in the cell surface membrane in order to assist in the attachment of adherent cells to extracellular matrices and of lymphocytes to other cells. The N-terminus of this protein contains elements for localization to cell-extracellular matrix junctions. The C-terminus contains binding sites for proteins such as beta-1-integrin, actin, and vinculin.
