Western blot of human T47D cells showing specific immunolabeling of the ~34 kDa cdc2 phosphorylated at Tyr 15 (Control, Lane 1). Treatment with EGF (30 ng per ml for 30 min) caused dephosphorylation of the Tyr 15 on cdc2 (Lane 2). The phosphospecificity of this labeling is also shown in lanes 3 and 4. These blots are identical to the control and EGF treated except that they were incubated in gamma-Ptase (1200 units for 30 min) before being exposed to the Anti-Phospho-Tyr 15 cdc2. The immunolabeling is completely eliminated by treatment with gamma-Ptase.Phosphospecificity is shown in lanes 3 and 4, where the immunolabeling is completely eliminated by blot treatment with lambda phosphatase (gamma-Ptase, 1200 units for 30 minutes).

The phosphorylation of beta-tubulins regulates microtubule dynamics during mitosis.
