Figure 1. Western blot analysis of IL18 using anti-IL18 antibody (PB9014). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: rat brain tissue lysates, . Lane 2: rat lung tissue lysates, . Lane 3: rat spleen tissue lysates. Lane 4: rat liver tissue lysates. Lane 5: rat testis tissue lysates. Lane 6: rat RH35 lysates. Lane 7: rat PC-12 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IL18 antigen affinity purified polyclonal antibody (Catalog # PB9014) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IL18 at approximately 22KD. The expected band size for IL18 is at 22KD.

背景 :
Interleukin (IL)-18, also called Interferon-gamma-inducing factor (IGIF), augments natural killer (NK) activity in spleen cells. The gene encodes a precursor protein of 192 amino acids and a mature protein of 157 amino acids. IL18 is a recently discovered cytokine that modulates both T helper type 1 (Th1) and Th2 responses. IL18 is a potent proinflammatory cytokine with potential atherogenic properties. It is highly expressed in the atherosclerotic plaques compared with control normal arteries and is localized mainly in plaque macrophages.
