Figure 1. Western blot analysis of IP3 receptor using anti-IP3 receptor antibody (PB9225). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. lane 1: Recombinant Human IP3 receptor Protein 0.5ng. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-IP3 receptor antigen affinity purified polyclonal antibody (Catalog # PB9225) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for IP3 receptor at approximately 45KD. The expected band size for IP3 receptor is at 45KD.

Figure 3. IHC analysis of IP3 receptor using anti-IP3 receptor antibody (PB9225). IP3 receptor was detected in paraffin-embedded section of Mouse Brain Tissue. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-IP3 receptor Antibody (PB9225) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Optimal dilutions should be determined by end users.

Protein Function :
Intracellular channel that mediates calcium release from the endoplasmic reticulum following stimulation by inositol 1,4,5- trisphosphate. Involved in the regulation of epithelial secretion of electrolytes and fluid through the interaction with AHCYL1 (By similarity). Plays a role in ER stress-induced apoptosis. Cytoplasmic calcium released from the ER triggers apoptosis by the activation of CaM kinase II, eventually leading to the activation of downstream apoptosis pathways (By similarity).

Inositol 1,4,5-trisphosphate receptor type 1, also known as IP3R or IP3R1, is a protein that in humans is encoded by the ITPR1 gene. It is mapped to 3p26.1. The product of the ITPR1 gene is predominantly enriched in cerebellar Purkinje cells but is also concentrated in neurons in the hippocampal CA1 region, caudate-putamen, and cerebral cortex. The ITPR1 gene encodes the inositol 1,4,5-triphosphate (IP3) receptor, an intracellular IP3-gated calcium channel that modulates intracellular calcium signaling. Upon stimulation by inositol 1,4,5-trisphosphate, this receptor mediates calcium release from the endoplasmic reticulum. Mutations in ITPR1 cause spinocerebellar ataxia type 15, a disease associated with an heterogeneous group of cerebellar disorders.
