Immunofluorescent analysis of Cyclin B1 (green) in HeLa cells either left untreated (left panel) or treated with 50nM Nocodazole (right panel) for 16 hours. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA (Product # 37525) for 15 minutes at room temperature. Cells were probed with a Cyclin B1 monoclonal antibody (Product # MA5-14327) at a dilution of 1:100 for at least 1 hour at room temperature, washed with PBS, and incubated with DyLight 488 goat anti-mouse IgG secondary antibody (Product # 35502) at a dilution of 1:400 for 30 minutes at room temperature. F-Actin (red) was stained with DyLight 554 Phalloidin (Product # 21834) and nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ArrayScan or ToxInsight Instrument at 20X magnification.

Immunohistochemistry was performed on formalin-fixed, paraffin-embedded human tonsil tissue. To expose target proteins, heat-induced antigen retrieval was performed by boiling tissue sections in 10mM sodium citrate buffer (pH 6) for 10-20 minutes. Immunoprecipitation of Cyclin B1 was performed on the Human Colon Carcinoma cell line LS174T. Antigen-antibody complexes were formed by incubating 500ul of LS174T whole cell lysate with 5ul of a Cyclin B1 monoclonal antibody (Product # MA5-14327). The immune complexes were captured using Protein A agarose, washed extensively, and eluted. Cyclins bind to and regulate the activity of the Cyclin dependent protein kinases (CDKs). Cyclin B1 or CCNB1 is a regulatory protein involved in mitosis. It is essential for the control of the cell cycle at the G2/M transition. Cyclin B1 complexes with p34 (cdc2) to form the maturation-promoting factor (MPF). Two alternative transcripts have been found, a constitutively expressed transcript and a cell cycle-regulated transcript, that is expressed predominantly during G2/M phase. Cyclin B1 is not ubiquitinated during G2/M phase, resulting in its steady accumulation during G2 phase, followed by abrupt APC dependent destruction at the end of mitosis. Destruction of Cyclin B1 is required for cell cycle progression. Cyclin B1 is overexpressed in various cancers, including breast, prostate, and non-small cell lung cancer.
