Recognizes human proteins of 92kD and 78kD, under non-reducing conditions, which are identified as pro (latent) and activated forms of matrix metalloproteinase 9 (MMP-9; also known as 92kD collagenase IV or gelatinase B). Also reacts with the complex (~200kD) of MMP-9 and tissue inhibitor of metalloproteinases-1 (TIMP-1). Shows no cross-reactivity with pro and active forms of MMP-2.

Immunohistochemistry: LS-B2259 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2259 was determined to be 5 ug/ml. Inhibition of MMP-9 Activity: 2-4 ug/ml. Immunoprecipitation: 2 ug/mg protein lysate. Use Protein G. Positive control: Conditioned, serum-free medium from (TPA-treated) human fibrosarcoma HT-1080 cells. The applications listed have been tested for the unmodified form of this product. Other forms have not been tested.
