Osteonectin (SPARC) [Mouse monoclonal antibody to Osteonectin (SPARC)]. This antibody reacts with human and bovine osteonectin as determined by Western Blotting, and cross-reacts with rabbit and porcine osteonectin as determined by ELISA. This antibody reacts with both bone and platelet derived osteonectin; it does not react with non-activated platelets, but it does react with thrombin-stimulated platelets, as determined by Flow Cytometry. treatment with 5mM EDTA for 30 minutes, as determined by Flow Cytometry.

Secreted protein acidic and rich in cysteine/osteonectin/BM40, or SPARC, is a matrix-associated protein that elicits changes in cell shape, inhibits cell-cycle progression, and influences the synthesis of extracellular matrix (ECM) (Bradshaw et al., 2003 [PubMed 12721366]).[supplied by OMIM]

Function: Appears to regulate cell growth through interactions with the extracellular matrix and cytokines. Binds calcium and copper, several types of collagen, albumin, thrombospondin, PDGF and cell membranes. There are two calcium binding sites; an acidic domain that binds 5 to 8 Ca2+ with a low affinity and an EF-hand loop that binds a Ca2+ ion with a high affinity.
