Accumulation of the amyloid- beta peptide (A beta ) in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. The beta -amyloid protein precursor (APP) is cleaved by beta -secretase, producing a soluble derivative of the protein and a membrane anchored 99-amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma -secretase to generate the 4 kD amyloid- beta peptide which is deposited in the brain.

This gene encodes a cell surface receptor and transmembrane precursor protein that is cleaved by secretases to form a number of peptides. Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy).
