This item specifically recognises human 5-lipoxygenase (5-LO, also known as arachidonate 5-lipoxygenase) when phosphorylated at Ser523. 5-LO is an enzyme that catalyses the synthesis of leukotrienes (LT's) from arachidonic acid (a polyunsaturated fatty acid), by the insertion of an oxygen into the arachidonic backbone. 5-LO requires calcium, ATP and the 5-LO-activating protein as cofactors for full activity. Leukotrienes are synthesised and secreted by leukocytes in response to immunologic or inflammatory stimuli, such as in asthma and allergic reactions. The biological effects of LT's include leukocyte chemotaxis, pulmonary airway constriction, mucous hypersecretion, vasoconstriction and increased vascular permeability. I recent years 5-LO has also been implicated in the regulation of cell proliferation and apoptosis and has been found in a broad variety of cancer cells.

Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use. This gene encodes a member of the lipoxygenase gene family and plays a dual role in the synthesis of leukotrienes from arachidonic acid. The encoded protein, which is expressed specifically in bone marrow-derived cells, catalyzes the conversion of arachidonic acid to 5(S)-hydroperoxy-6-trans-8,11,14-cis-eicosatetraenoic acid, and further to the allylic epoxide 5(S)-trans-7,9-trans-11,14-cis-eicosatetrenoic acid (leukotriene A4). Leukotrienes are important mediators of a number of inflammatory and allergic conditions. Mutations in the promoter region of this gene lead to a diminished response to antileukotriene drugs used in the treatment of asthma and may also be associated with atherosclerosis and several cancers.
