This kit recognizes natural and recombinant Rat IGFBP-3. No significant cross-reactivity or interference between Rat IGFBP-3 and analogues was observed.

Sensitivity: The minimum detectable dose of Rat IGFBP-3 is 46.875pg/mL (The sensitivity of this assay, or lowest detectable limit (LDL) was defined as the lowest protein concentration that could be differentiated from zero).

Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Rat IGFBP-3 concentrations in serum, plasma and other biological fluids.

Principle of the assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to IGFBP-3. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for IGFBP-3 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain IGFBP-3, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of IGFBP-3. You can calculate the concentration of IGFBP-3 in the samples by comparing the OD of the samples to the standard curve.

This gene is a member of the insulin-like growth factor binding protein (IGFBP) family and encodes a protein with an IGFBP domain and a thyroglobulin type-I domain. The protein forms a ternary complex with insulin-like growth factor acid-labile subunit (IGFALS) and either insulin-like growth factor (IGF) I or II. In this form, it circulates in the plasma, prolonging the half-life of IGFs and altering their interaction with cell surface receptors. IGFBP3: IGF-binding proteins prolong the half-life of the IGFs and have been shown to either inhibit or stimulate the growth promoting effects of the IGFs on cell culture.
