Anti-Src antibodies (Src-101AP, Src-112AP) The Src family of protein tyrosine kinases are important regulators of cell growth and differentitation of eukaryotic cells. The src protien has a SH3, SH2 and a kinase domain. SH2 domain containing proteins play a key role in the process of intracellular transmission of signalling events initiated at the cell surface. As a pre-requisite in the fulfillment of this function, these proteins bind to a variety of phospho-tyrosine (pY) containing target molecules. The SH3 domainThe Src family cosist of several protein kinases including Src, Lyn, Fyn, Yes, Lck, Blk, Hck, etc. (1). The src activity is modulated in opposite directions by phosphorylation of tyrosie residues 516 and 527. The phosphorylation of Tyr-416 resulted in activation of src kinase actvity, while a negative regulation of the cellular Src tyrosine kinase (pp60c-src) is mediated through the phosphorylation of a C-terminal tyrosine residue, Tyr-527 that is located 11 residues C-terminal from the end of the kinsase domain (2, 3). Current models predict that inhibition of c-Src kinase activity results from an interaction of phosphorylated Tyr-527 with the amino terminal SH2 domain. One of the substrate for c-src kinase is c-fgr, a protooncogene product. The c-scr protein is a approximately 57kDa protein. The Anti-scr-selective antibody was generated against 2 peptides (DSGGFYITSR TQF) and (LIEDNEYT ARQGAC) from the tyrosine kinase domain that is unique to c-src protein. The phospho-sopecific affinity purified mono-epitope specific antibodies were selected on multiple column of dephospho and phospho-peptide immobilized antigen chromatograpgy. The affinity purified antibodies strongly label a 57kDa c-Src protein in our western blot positive control samples. MyBioSource will also conjugate antibodies with fluorescent probes upon request at extra charge. MyBioSource also provides antibodies against proteins that are involved in ACK1 signalling including ACK2, phosphor-ACK1, Herceptins (Erbs), bone morphogenetic proteins and others. MyBioSource employs cyclic peptide methodology for generating antibodies, which results in higher titer and specificity (6). MyBioSource, will also provide Western blot positive controls for phospho and non-phospho src proteins in ready-to-use buffer for easy identification of respective proteins. This gene encodes three main isoforms that differ in activities and subcellular location. While all three are adapter proteins in signal transduction pathways, the longest (p66Shc) may be involved in regulating life span and the effects of reactive oxygen species. The other two isoforms, p52Shc and p46Shc, link activated receptor tyrosine kinases to the Ras pathway by recruitment of the GRB2/SOS complex. p66Shc is not involved in Ras activation.
