Recombinant CD4 was lyophilized from a 0.2 um filtered solution in PBS, pH 7.4.

Stable for at least 2 year of receipt at -20 degree C. Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

Domain: IG. Host Note: Optimized DNA sequence encoding Human CD4 extracellular domain was expressed in HEK293 cells. Activity: Measured by the ability of the immobilized protein to support the adhesion of HeLa human cervical epithelial carcinoma cells. Biological Activiy: The activity was tested by immobilized recombinant CD4(1ug/ml) ability to support adhesion of HeLa cells (5x10^4) cells/well). 40-80% will adhere after 60 mins at 37 deg. C. Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1mg/ml. This solution can then be diluted into other buffers.

Endotoxin: Endotoxin content was assayed using a LAL gel clot method. Edotoxin level was found to be less than 0.1ng/ug (1EU/ug). Molecular Weight: Human soluble CD4 is generated by the protolytic removal of the extracellular domain and propeptide. The molecule has a calculated molecular mass of approximately 40 kDa. Recombinant CD4 is a disulfide-linked monomer protein consisting of 376 amino acid residue subunits, and migrates due to glycosylation as an approximately 46 kDa protein under non-reducing and reducing conditions in SDS-PAGE.

This gene encodes a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus. This gene is expressed not only in T lymphocytes, but also in B cells, macrophages, and granulocytes. It is also expressed in specific regions of the brain. The protein functions to initiate or augment the early phase of T-cell activation, and may function as an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system.
