The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this protein can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

Recombinant human EBV EBNA1 antigen is suitable for ELISA and Western blots, excellent antigen for detection of EBV with minimal specificity problems.

Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers. Molecular Weight Note: Recombinant EBV EBNA-1 encodes the immunodominant sequences of theEpstein-Barr Virus nuclear antigen 1 protein. Recombinant EBV EBNA1 contains an N terminal GST purification tag and migrates as an approximately 46 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.

EBNA1 is the only EBV protein consistently expressed in all proliferating infected cells and plays several important roles in EBV latent infection, including the initiation of EBV DNA replication, the mitotic segregation of the EBV genomes, and transcriptional activation of other EBV latency proteins. several pieces of evidence suggest that EBNA1 plays a direct role in cellular transformation by EBV. First, EBNA1 is expressed in all EBV-associated tumors and is the only viral protein expressed in some of these tumors. Second, transgenic mice expressing EBNA1 develop malignant B cell lymphomas. Third, the expression of EBNA1 in Hodgkin cells enhances their ability to form tumors in nonobese diabetic-SCID mice. Fourth, EBV genomes lacking the EBNA1 gene are several thousand-fold less efficient at B cell immortalization than EBV genomes expressing EBNA1. Fifth, interference with EBNA1 function in Burkitt's lymphoma cells by overexpression of the EBNA1 DNA binding domain increased cell death, suggesting that EBNA1 normally provides a survival function for these cells.

Function: Plays an essential role in replication and partitioning of viral genomic DNA during latent viral infection. During this phase, the circular double-stranded viral DNA undergoes replication once per cell cycle and is efficiently partitioned to the daughter cells. EBNA1 activates the initiation of viral DNA replication through binding to specific sites in the viral latent origin of replication, oriP. Additionally, it governs the segregation of viral episomes by mediating their attachment to host cell metaphase chromosomes. Also activates the transcription of several viral latency genes.
