Tumor Necrosis Factor-alpha (TNF-?) is a multifunctional pro-inflammatory cytokine, mainly secreted by activated macrophages. It is implicated with a variety of biological procedures including systemic inflammation, cell proliferation, apoptosis, lipid metabolism, and coagulation. The pleiotropic attribute of TNF-? regulation is associated with its ability to trigger multiple signalling pathways through its receptors, TNFR1 (p55) and TNFR2 (p75). TNF-? not only contributes to the immune response to bacterial, fungal, viral and parasitic invasions, but also functions in tissue remodeling, autoimmune-diseases and the necrosis of specific tumors. TNF-? hyper-expression in response to some bacterial components such as LPS can cause life threatening septic shock. Recombinant TNF-?, in combination with chemotherapy, has been applied for treatment of soft sarcomas, melanomas and other irresectable tumors. Anti-TNF-? therapy has been used for treatment of rheumatoid arthritis.

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer.
