This assay has high sensitivity and excellent specificity for detection of bovine CCL2. No significant cross-reactivity or interference between bovine CCL2 and analogues was observed.

Typical Testing Data/Standard Curve (for reference only)

Samples: Serum, Plasma, Tissue Homogenates. Assay Type: Quantitative Sandwich. Detection Range: 78 pg/ml-5000 pg/ml. Sensitivity: 19.5 pg/ml.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CCL2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CCL2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCL2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4. Function: Chemotactic factor that attracts monocytes and basophils but not neutrophils or eosinophils. Augments monocyte anti-tumor activity. Has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis or atherosclerosis. May be involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis.
