This assay has high sensitivity and excellent specificity for detection of PRKCE . No significant cross-reactivity or interference between PRKCE and analogues was observed.

Store at 4 degree C if kit is to be used within 1 week. Stable for 6 months (if micro ELISA Plate, Lyophilized Standard and Concentrated Biotinylated Detection Protein stored at-20 degree C. Other components at 2-8 degree C). Stable for 12 months (if the entire kit is stored at-20 degree C).

Intra-assay Precision: Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level PRKCE were tested 20 times on one plate, respectively. Intra-Assay: CV<8%. Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- PRKCE antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- PRKCE antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the PRKCE amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of PRKCE can be calculated.

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been shown to be involved in many different cellular functions, such as neuron channel activation, apoptosis, cardioprotection from ischemia, heat shock response, as well as insulin exocytosis. Knockout studies in mice suggest that this kinase is important for lipopolysaccharide (LPS)-mediated signaling in activated macrophages and may also play a role in controlling anxiety-like behavior.
