Fluorescent image of A549 cell stained with SRC Antibody. A549 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with SRC primary antibody (1:25, 1 h at 37 degree). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-mouse antibody (green) was used (1:400, 50 min at 37 degree).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree).SRC immunoreactivity is localized to Cytoplasm significantly.

Function: Non-receptor protein tyrosine kinase which is activated following engagement of manydifferent classes of cellular receptorsincluding immune response receptors,integrins and other adhesion receptors,receptor protein tyrosine kinases, G protein-coupled receptors as well as cytokinereceptors. Participates in signaling pathwaysthat control a diverse spectrum of biologicalactivities including gene transcription,immune response, cell adhesion, cell cycleprogression, apoptosis, migration, andtransformation. Due to functional redundancybetween members of the SRC kinase family,identification of the specific role of each SRCkinase is very difficult. SRC appears to be oneof the primary kinases activated followingengagement of receptors and plays a role inthe activation of other protein tyrosine kinase(PTK) families. Receptor clustering ordimerization leads to recruitment of SRC tothe receptor complexes where itphosphorylates the tyrosine residues withinthe receptor cytoplasmic domains. Plays animportant role in the regulation ofcytoskeletal organization throughphosphorylation of specific substrates such as AFAP1. Phosphorylation of AFAP1 allows theSRC SH2 domain to bind AFAP1 and tolocalize to actin filaments. Cytoskeletalreorganization is also controlled through thephosphorylation of cortactin (CTTN).
