Anti-PhosphoTyrosine antibody is suitable for use in ELISA and western blotting assays.  The antibody reacts specifically with phosphotyrosine  and shows minimal reactivity by ELISA and western blotting with proteins containing phosphothreonine or phosphoserine residues.  The antibody reacts with free phosphotyrosine, phosphotyrosine conjugated to carriers such as thyroglobulin or BSA, and detects the presence of phosphotyrosine in proteins of both unstimulated and stimulated cell lysates.  Although not tested, this antibody is likely functional in RIA, flow cytometry and immunohistochemistry.

Anti-PhosphoTyrosine was prepared from monospecific antiserum by immunoaffinity chromatography using phosphorylated tyrosine peptide coupled to agarose followed by conjugation to biotin and subsequent purification.   Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit IgG and anti-Biotin.  Reactivity is specific for phosphotyrosine and minimal cross reactivity is observed against phosphothreonine or phosphoserine.

Store Anti-pY at -20° C prior to opening.  This product is stable for several weeks at 4° C as an undiluted liquid.  Dilute only prior to immediate use.  For extended storage aliquot contents and freeze at -20° C or below.  Avoid cycles of freezing and thawing.

Anti-PhosphoTyrosine detects phophorylated Tyrosines. Tyrosine is one of the 22 amino acids that are used by cells to synthesize proteins. Tyrosine has a phenol group. It functions as a receiver of phosphate groups that are transferred by way of protein kinases. Phosphorylation of the hydroxyl group changes the activity of the target protein.
