This intronless gene encodes a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. The product of this gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. This gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT). Applications: Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence, Flow Cytometry and ELISA. Other applications have not been tested. Recommended Dilution: Western Blot: 1:200-1:2000 Immunohistochemistry (FFPE): 1:50-1:100 Immunofluorescence: 1:100 Flow Cytometry: 1:10-1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Fluorescent image of A549 cell stained with 031255. A549 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with SOX2 primary antibody (1:25, 1 h at 37ºC). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-mouse antibody (green) was used (1:400, 50 min at 37ºC). Cytoplasmic actin was counterstained with Alexa Fluor® 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37ºC). SOX2 immunoreactivity is localized to Nucleus significantly.
