MYBPC3 encodes the cardiac isoform of myosin-binding protein C. Myosin-binding protein C is a myosin-associated protein found in the cross-bridge-bearing zone (C region) of A bands in striated muscle. MYBPC3, the cardiac isoform, is expressed exclussively in heart muscle. Regulatory phosphorylation of the cardiac isoform in vivo by cAMP-dependent protein kinase (PKA) upon adrenergic stimulation may be linked to modulation of cardiac contraction. Mutations in MYBPC3 are one cause of familial hypertrophic cardiomyopathy. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications have not been tested. Recommended Dilution: Western Blot: 1:1000-1:2000 Immunohistochemistry (FFPE): 1:25 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunohistochemistry of MYBPC3 in paraformaldehyde-fixed, paraffin-embedded sections of human heart tissue sections using 038715. Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5hrs at RT; antigen retrieval was by heat mediation with a citrate buffer (pH 6). Samples were incubated with 038715 (1:25) for 1hr at 37°C. A undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.
