Recognizes a polypeptide which is identified as insulin, a 51aa polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as beta-cell and insulinoma marker. Applications: Suitable for use in Immunofluorescence, Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 0.5-1ug/10e6 cells Immunofluorescence: 1-2ug/ml Immunohistochemistry (FFPE): 0.1-0.2ug/ml for 30 minutes at RT No special treatment is required for staining of formalin/paraffin tissues. Optimal dilutions to be determined by the researcher. Positive Control: MIA PaCa-2 cells or Pancreas Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
