Notch1 is a transmembrane protein functioning in development and the determination of cell-fate. During maturation, the notch molecule is cleaved by a furin-like convertase at its extracellular domain. Upon binding to a ligand such as D1, or upon extracellular calcium depletion, the carboxy-terminal notch1 fragment is released and further cleaved between Gly1743 and Val1744. The resulting activated cytosolic fragment translocates to the nucleus where it activates transcription. Notch4 is primarily expressed in endothelial cells and is processed similarly to Notch1. The mouse Notch4 gene (also known as Int-3) is a frequent target in Mouse Mammary Tumour Virus (MMTV)-induced mammary tumors. The intracellular domain of Notch4 binds to Smad3 inhibits the TGF- beta signaling. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000 Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
